I need to replace my already flashed Hitachi 78FK with a new drive. Do I need to insert the original key BEFORE I flash with the hacked FW, or can I just flash it with the hacked FW without inserting the driev key?

You need to insert the drive key and flash those sectors. Then apply the hack in 360FWTB, which will update your master checksum.
