Firstly you have post in the X3 Bios Flashing Support area. The DUOX is not an XECUTER 3 chip so do not try an Xecuter 3 Bios.
i am not sure of the flashing process for a DUOX chip as I have only ever used Xecuter branded chips. You may find more help regarding flashing on the DUOCHIP website:
http://www.duomodchip.com/english/install.htm - JUST CHECKED AND THERE IS A TUTORIAL FOR FLASHING THE CHIP ON THAT WEBSITE!!!! It seems that it is basically the same as flashing an Xecuter chip but just to be safe use their tutorial.
Also after doing some reading about the chip it appears that it has a 512k flash area so the best bios for you to use would be the Xecuter 2 5035 bios. Be careful though as there are two versions of this bios. X2_5035_old.bin(for 1.0 - 1.5 boxes) and X2_5035_16(for 1.6 or 1.6b boxes).
And in answer to your final question yes you will need a dash installed to use any new homebrew software. As you are new to this I would start with EVOX dash as it is very easy to configure. When you are more confident try switching to something like avalaunch, Unleash or XBMC.
